Design Notes
This is a subpart which will be integrated for full functionality. The cis repressor is a regulatory element which sequesters the RBS of the mRNA transcript for the gene of interest and thus suppresses expression of that gene product. It is located downstream of the promoter to the regulated gene and shortly (~6bp upstream of the start codon). In this part, the YFP gene is repressed by the cis element and is under the control of the Ptet promoter. This part will be integrated with part S03724, which contains the TetR gene under the control of a constitutive promoter. TetR represses the Ptet promoter and thus allows for an inducible system for expressing the gene of interest.
